Poltekkes Kemenkes Semarang sejak tahun 2007 berkomitmen untuk melaksanakan sistem manajemen mutu dalam mengawal mutu pendidikan yang mengarah pada peningkatan mutu lulusan. Komitmen tersebut terjustifikasi dengan diraihnya sertifikasi penjaminan mutu berbasis ISO 9001:2000 pada tahun 2009 dan dipudate dengan ISO 9001:2008 pada tahun 2010. Proses penjaminan mutu selalu dikawal baik melalui Audit Mutu Internal (AMI) yang dilaksanakan setiap periode Mei dan November oleh audior-auditor internal yang telah mendapatkan pelatihan dan lulus sebagai auditor. Di samping itu dilakukan pula audit mutu eksternal dai badan sertifikasi PT. SGS Indonesia.

Proses sertifikasi penjaminan mutu berbasis ISO yang disertifikasi oleh PT. SGS Indonesia berlaku untuk tiga tahun, sehingga pada tahun 2012 dilakukan re-sertifikasi pertama dan pada tahun 2015 di lakukan re sertifikasi kedua. Proses surveilance audit yang dilakukan oleh PT SGS sejak tahun 2015 dilakukan sekali dalam setahun setiap bulan Maret dengan cakupan 75 % seluruh Program Studi.

Pada hari Kamis tanggal 16 Maret 2017 telah dilakukan Audit Mutu Ekternal (AME) dari PT SGS pada Prodi D III Rekam Medis dan Informasi Kesehatan Poltekkes Kemenkes Semarang. Kegiatan berjalan tertib yang dilakukan oleh Auditor Ibu Renata Hutapea. Kegiatan dilakukan dari pukul 13.00 sampai dengan pukul 16.00, di lingkungan kampus Jurusan RMIK Poltekkes Semarang dan disambut secara langsung oleh Ketua Jurusan RMIK Bapak Edi Susanto, SH, S.Si, M.Kes.
